β-Actin Antibody

β-Actin (also known as muscle actin or cytoskeletal actin) is composed of 375 amino acids with a molecular weight of approximately 42-43kDa. It is widely distributed in the cytoplasm of most tissues and cells, making it a commonly used reference protein for detecting changes in protein expression levels. β-Actin is recognized as an internal control due to its stable expression levels across different tissues and cells, although its expression may be lower in specific cases such as adipose tissues and cells.


GAPDH Antibody

GAPDH (Glyceraldehyde 3-phosphate dehydrogenase) is a key enzyme involved in glycolysis, composed of four subunits of 30-40kDa each, totaling a molecular weight of 146kDa with detection bands around 36kDa. GAPDH gene is highly expressed in almost all tissues and cells, maintaining relatively constant levels within the same cell or tissue type and is minimally influenced by external stimuli. It is widely used as an internal reference for WB protein standardization. However, conditions like hypoxia and diabetes can increase GAPDH expression in specific cells and tissues.

Data

Perform WB validation analysis using HEK-293, Hepal-6, PC12, CHO-K1, and MDCK cells. Electrophoresis at 120V after loading, followed by membrane transfer at 300mA for 1.5 hours. Block with 5% skim milk powder for 2 hours post-transfer. Incubate with β-Actin antibody (Catalog#GM-77430AB) at 1:1000 dilution overnight at 4°C on a shaker. Wash three times with 1×TBST, incubate with secondary antibody at room temperature for 2-3 hours on a shaker, followed by three washes with 1×TBST before developing.

Use HEK-293, Jurkat, PC12, WSL, Hepal-6, and MDCK cells for WB validation analysis. After loading the samples, conduct electrophoresis at 120V, followed by transfer at 300mA for 1.5 hours. Post-transfer, block with 5% skim milk powder for 2 hours. Incubate overnight at 4°C with GAPDH antibody (Catalog#GM-77904AB) at 1:1000 dilution, wash thrice with 1×TBST before incubating at room temperature with secondary antibody for 2-3 hours, followed by three washes with 1×TBST prior to imaging.
