Interleukin-11 (IL-11) is an important hematopoietic cytokine expressed in the central nervous system and gastrointestinal tract. Its hematopoietic effects include stimulation and proliferation of primitive stem cells. IL-11 acts synergistically with other cytokines to support the proliferation and differentiation of hematopoietic stem cells of all lineages. Additionally, in animal models of acute and chronic inflammation, IL-11 has been found to be associated with anti-inflammatory and mucosal protective effects, such as mucositis, inflammatory bowel disease, arthritis, and autoimmune arthropathy.
IL-11 signals by binding to the IL-11 receptor-α (IL-11Rα) and gp130 complex. IL-11Rα, similar to other members of the IL-6 receptor family, consists of the IL-11-specific α chain, while gp130 subunit is a common receptor for the β chain subunit. Binding of IL-11 to IL-11Rα triggers homodimerization of the gp130 subunit and formation of the IL-11 receptor complex. Signaling is transmitted through the gp130 receptor and initiates downstream gene transcription via the JAK/STAT pathway.
Genomeditech's H_IL-11 Reporter 293 Cell Line is a luciferase reporter cell line constructed based on the STATs signaling pathway. Upon IL-11 binding to IL-11Rα and GP130 receptors, the signaling pathway is activated, leading to the expression of firefly luciferase. Luciferase readout reflects the activation effect of the signaling pathway, thus can be used for in vitro evaluation of IL-11 related drugs.
