Background KLK2 is a membrane-bound trypsin-like serine endopeptidase that is specifically expressed in prostate epithelial cells and prostate cancer cells under the regulation of and Rogen receptor (AR) and acts synergistically with prostate-specific antigen (PSA) , which is essential for the development of prostate cancer, activation of PSA function by enzyme digestion is a key regulator of PSA biological activity. The positive rates of KLK2 in localized prostate cancer (LPC) , metastatic hormone-sensitive prostate cancer (MHSPC) and metastatic castration-resistant prostate cancer (mCRPC) were 91% , 81% and 76% , respectively, the expression of PSMA is strictly specific to prostate tissue, and there is almost no signal in normal tissues (such as salivary gland and kidney) , which greatly reduces the risk of off-target. KLK2 is not only involved in tumor cell proliferation, angiogenesis and microenvironment remodeling, but also closely related to tumor invasiveness and castration resistance, and its high expression level is positively correlated with the stage of disease progression, and the expression level of KLK2 is positively correlated with the stage of disease progression, and it is not affected by AR pathway inhibitors, which provides an alternative treatment option for patients with AR pathway inhibitor resistance.
