Background Interleukin-18 (IL-18) is a pro-inflammatory cytokine that plays a pivotal role in the immune response. It is produced as an inactive precursor and activated by proteolytic processing, enabling it to promote the production of interferon-gamma (IFN-γ) and amplify Th1 immune responses. IL-18 works in concert with IL-12 to enhance natural killer (NK) cell activity and T-cell activation, contributing to defenses against intracellular pathogens. Beyond its protective roles, dysregulated IL-18 signaling has been implicated in inflammatory and autoimmune conditions, making it a target for therapeutic intervention in diseases such as rheumatoid arthritis, inflammatory bowel disease, and metabolic disorders. Owing to its broad involvement in inflammation, IL-18 is studied as both a biomarker of disease activity and a potential candidate for cytokine-modulating therapies.
