IL-2 (interleukin-2) is a cytokine produced by T cells and plays an important role in the immune system. The IL-2 promoter is located in the upstream region of the IL-2 gene and contains multiple important transcription factor binding sites, including NFAT (nuclear factor activated T cells), AP-1 (activated protein-1), and NF - κ B (nuclear factor kappa B). When T cells are activated, these transcription factors bind to the IL-2 promoter, promoting the transcription and expression of IL-2. The activity of IL-2 promoter is regulated by multiple signaling pathways, such as T cell receptor (TCR) signaling, co stimulatory signaling (e.g. CD28), and other cytokine signaling.
The H_IL2 Promoter Reporter Jurkat Cell Line of Jiman Biotechnology is a stable cell line constructed by placing the IL-2 promoter upstream of the luciferase reporter gene and transfecting it into Jurkat cells. Jurkat cells naturally express TCR/CD3 complexes and CD28 co stimulatory receptors. Research drugs can activate these co stimulatory signaling pathways, activate IL-2 promoter driven luciferase expression, and thus be used for in vitro activity evaluation of drugs targeting T cell activation direction.
