Description

ADCP, or Antibody-Dependent Cell-Mediated Cytotoxicity, refers to the process where immune cells expressing Fc receptors engulf target cells by recognizing the Fc portion of antibodies binding to the target cell. Nowadays, the ADCP mechanism is utilized for detecting and assessing the potency of antibodies. When the antibody's Fab segment binds to the target antigen on the cell surface, and the antibody's Fc segment simultaneously binds to the FcγRIIb receptor on effector cells (mainly macrophages), crosslinking occurs, activating the ADCP pathway leading to target cell engulfment. In the traditional ADCP biological activity assay, macrophages from donors are used as effector cells, which exhibit considerable response variability, making them difficult to prepare and leading to high background readings.

The Genomeditech's ADCP FcγRIIb Jurkat Effector Cell Line uses engineered Jurkat cells as effector cells. Through clever vector design and a third-generation lentivirus reporter gene system, it stably expresses human FcγRIIb receptor and firefly luciferase driven by a transcription factor to quantify the biological activity of antibodies in the ADCP mechanism activated through the NFAT pathway, with luciferase production, and the luciferase activity in effector cells quantified through bioluminescence readings. This method provides high signal values with low background levels. Custom ADCP cell lines and ADCP activity testing services are also available.
